Updating and Editing Calibration Files in Chromperfect

Welcome to this Chromperfect training guide. In this post, we’ll walk through working with a Chromperfect calibration files inside the Analysis program, covering how to update, adjust, create, and manage components. These tools help ensure accurate quantification and smooth operation of your chromatography workflows. Created using Chromperfect version 8 but still relevant in later versions.


Updating the Chromperfect Calibration File

Instead of always creating and processing a bound file, Chromperfect allows you to update the calibration file directly from a raw file:


  • Load a raw file.

  • Adjust the integration if necessary.

  • Go to Components → Update Calibration File.


The form that appears resembles the calibration file editor. Since the integration data is already in memory, no bound file name is needed. When updating several levels, it’s best to complete all changes before printing an update report. An additional checkbox allows you to control whether the calibration file is saved.


Adjusting the Calibration File

When a raw file is integrated, the Analysis program matches peaks to components using retention times and window widths defined in the calibration file. If identifications are incorrect, you can:


  • Modify retention time or window width.

  • Define new components.

  • Delete unwanted components (with some exceptions).


While adjustments can be made here, the Calibration Editor remains the best tool for setting default retention references, internal standards, and large-scale edits.


Creating a New Calibration File

To start fresh:


  • Go to Components → Make New Calibration File.

  • Confirm whether to overwrite the file in memory.


This clears the current calibration, but existing files remain intact until explicitly overwritten.


Editing the Component List

Access the full list of components via Components → Show Component List. Here, you can:


  • Edit names, retention times, and widths.

  • Insert new components (highlighted in yellow).

  • Delete components (except defaults or references).


Changes are highlighted until applied, and the chromatogram updates automatically.


Adding a New Component

  1. Go to Components → Modify Components → Add.

  2. Click on the chromatogram where the peak appears.

  3. Enter the component name, retention time, and window width.

  4. Specify quantification (area or height).

Temporary labels and vertical lines show the new search window, which can be dragged to adjust. Once confirmed, the chromatogram reintegrates with the new component included.


Editing an Existing Component

There are two methods:

  • Numeric Edit: Modify component values in a form.

  • Graphic Edit: Click and drag directly on the chromatogram to adjust retention time and window width.

Both methods reintegrate the chromatogram after confirmation.


Deleting Components

Use Components → Modify Components → Delete. Select the component on the chromatogram, confirm, and the reintegrated plot will remove it. Components defined as defaults, references, or internal standards cannot be deleted.


Nudging Components

For shifting multiple components by the same amount:

  1. Go to Components → Modify Components → Nudge.

  2. Drag a region on the chromatogram.

  3. Move the group horizontally to adjust retention times consistently.

This is particularly useful when retention times shift across an entire run.
